In my experience, there is no place to select to use your account'settings built up credit when making a purchase, but Amazon applies it when possible. So, if you add all of the cards to your account and make your purchase, then the payment method you choose should only be charged if your account doesn't have enough credit to fully cover it.
When in doubt, though, you can always chat with customer service. They are quite pleasant to deal with and can get you a definite answer.

Yeah, it's kind of weird, but it forces you to not forget about using them. But then instead of using them on a specific purchase (I'm going to use that gift card for...) then you end up using them for dumb stuff like light bulbs.
